Growth hormone secretagogue research

Growth hormone is released in pulses from somatotroph cells in the anterior pituitary, under opposing control from GHRH, which stimulates release, and somatostatin, which suppresses it. Ghrelin receptor agonists act through a third, independent pathway. Because the two stimulatory routes converge on the same cell through different receptors, compounds from each class are commonly studied together.

Selectivity is the central variable in this category. Early secretagogues raised growth hormone but also cortisol and prolactin, confounding any downstream measurement. Separately, hGH fragments isolate one region of the intact hormone. Studying the C-terminal lipolytic region alone allows adipocyte effects to be examined without the IGF-1 response that full-length growth hormone produces — a clean separation that is impossible with the intact molecule.

Practical notes for this category

Pulsatility matters in study design. Compounds with short half-lives model discrete pulses; long-acting versions produce sustained elevation, which is a physiologically different stimulus and produces different receptor desensitisation behaviour. Reading a product listing

Choosing between similar compounds

Several groups in the catalogue contain compounds that look interchangeable and are not. Among the incretins, semaglutide is a single GLP-1 receptor agonist, tirzepatide adds GIP receptor activity, and retatrutide adds glucagon receptor activity on top of both.
